Incorporation of Tritiated Cytidine into Ribonucleic Acid by Isolated Pea Nuclei

چکیده
A new method for the isolation of the nuclei from plant tissue has been devised. This method consists in passing the plant tissue through a set of spring-loaded, counter-rotating rollers and collecting the liberated nuclei in sucrose solution containing calcium ions. A semi-automatic machine which couples the rollers with a special tissue chopping device has been constructed. It has been shown that nuclei obtained in this way actively incorporate cytidine-H(3) into RNA. This incorporation is increased in the presence of nucleoside triphosphates and an energy-regenerating system.

have been prepared and studied for possible incorporation into polynucleotides. Arab inosylcytosine mono-, di-, and triphosphates were synthesized by both chemical and enzymatic methods. Labeled and unlabeled arabinosylcytosine nucleotides were tested for their capacity to serve as substrates for purified polynucleotide phosphorylase, deoxyribonucleic acid-dependent ribonucleic acid polymerase,...

There is still controversy concerning the primary site of RNA synthesis in the cell.' When tissues are radioautographed after administration of radioactive inorganic phosphorus or radioactive RNA2 precursors such as tritiated uridine or cytidine, it is commonly observed that radioactivity appears in the nucleus first and in the cytoplasm later. It has furthermore been shown that isolated nuclei...

In spite of recent notable achievements in the isolation and purification of enzymes capable of synthesizing polynucleotides (l-3), the identity of those enzymes responsible for the synthesis of ribonucleic acid in mammalian cells has not been established. The nucleoside diphosphates of the four bases of ribonucleic acid can be polymerized into a polynucleotide which resembles a natural ribonuc...

متن کاملThe incorporation of nucleotides into amino acid transfer ribonucleic acid. II. Evidence for separate enzymatic sites for incorporation of adenosine 5'-monophosphate and cytidine 5'-monophosphate.
The preceding paper described the purification and some of the properties of an enzyme present in rabbit muscle extracts that catalyzes the incorporation of adenylic acid from adenosine triphosphate into the terminal position of amino acid transfer ribonucleic acid obtained from yeast (1). The data presented in this paper are concerned with the incorporation of cytidine 5’-monophosphate into th...
